This strain is genotyped using generic Venus primers. There is no allele specific assay available at this time.
These K5-Venus transgenic mice express the yellow fluorescent protein Venus variant, under the control of the bovine keratin 5 promoter, and have applications in lineage tracing of the epithelial progenitor cell population in the developing submandibular gland and sublingual gland.

Keratin 5, also known as Cytokeratin-5 protein, is a type II cytokeratin specifically expressed in the basal layer of the epidermis, and is a basal epithelial cell marker in adult mouse salivary glands. These transgenic mice express the Venus variant of yellow fluorescent protein under the control of the bovine, keratin 5, promoter.
Fluorescence is detected in the epithelial progenitor cells in the end buds and ducts of the developing submandibular gland and sublingual gland. The Donating Investigator reports transgene expression was detected in all epithelial tissues including: skin, tongue, oral cavity lung, and forestomach.
Transgene expression mimics the endogenous mouse Krt5 gene expression pattern.
During backcrossing, the Y chromosome may not have been fixed to the C57BL/6 genetic background. Mice that are homozygous for the transgene are viable and fertile on the FVB background. The Donating Investigator reports that homozygotes exhibit a stronger Venus YFP expression compared to hemizgyotes.
A transgenic construct containing the Venus variant of yellow fluorescent protein under the control of the 5.4 kb bovine, keratin 5 promoter (including 5’ regulatory and promoter regions from -5,300 bp to +138 bp), and a polyadenylation signal, was injected into fertilized FVB/N blastocysts. Founder line 1 was subsequently established. The mice were then backcrossed to C57BL/6 for 3 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6 genetic background.
Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

When maintaining a live colony, hemizygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). The Donating Investigator has not attempted to make the strain homozygous on the C57BL/6 background.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
