Lmnaflox/flox mutant mice possess loxP sites flanking exon 2 of the lamin A (Lmna) gene. Lamin-A is part of the nuclear lamina seen in increasing levels of expression as development progresses. It is required for normal development of peripheral nervous system and skeletal muscle and plays an important role in nuclear assembly, chromatin organization, nuclear membrane and telomere dynamics. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues. Since different Lamin-A isoforms exist, this strain was developed to produce offspring that lack the majority of Lamin-A isoforms.
A targeting vector was designed to insert a loxP site upstream of exon 2, and a frt-flanked puromycin resistance (puro) cassette followed by a second loxP site, downstream of exon 2 of the lamin A (Lmna) gene. The construct was electroporated into (C57BL/6 x 129S4/SvJae)F1-derived V6.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric males were bred with CD1 females. Offspring were bred to B6;SJL-Tg(ACTFLPe)9205Dym/J transgenic mice (Stock No. 003800) to delete the puro cassette, and progeny were crossed to remove the Flp-expressing transgene. The resulting Lmnaflox/flox colony was maintained on a mixed background. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

Molecular Note | A loxP site was inserted upstream of exon 2. An FRT-flanked puromycin resistance cassette with a 3' loxP site was inserted downstream of exon 2. Flp-mediated recombination removed the selection cassette and left exon 2 floxed. |
When maintaining a live colony, homozygous mice may be bred together.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
