This spontaneous mutant is valuable in the assessment of craniofacial, hearing, and fertility defects related to Prkra and as part of a phenotypically diverse allelic series.

Mice homozygous for the recessive little ear 3 Jackson mutation can be recognized by 2 weeks of age by their smaller ear pinnae. They also have a smaller overall body size, are infertile, and at eight weeks of age were found to have a severe hearing deficit. This is a distinct phenotype from that of the Prkratm1Wsmay targeted deletion, which causes complete lethality by embryonic day 3.5.
The little ear 3 Jackson mutation arose spontaneously in the strain B6.B10ScN-Tlr4lps-del/JthJ strain at The Jackson Laboratory in 2010. The Tlr4lps-del mutation was bred out of this mutant subline by backcrossing once to C57BL/6J and selectively sibling intercrossing thereafter.

Strain of Origin | B6.B10ScN-Tlr4lps-del/JthJ |
Chromosome | 2 |
Molecular Note | This spontaneous C to T transition at Chromosome 2 position 76,643,218 bp (GRCm38), right after exon 3, likely disrupts splicing of the pre-mRNA. |
Because homozygotes are infertile this strain is maintained by intercrossing heterozgyotes.

The Jackson Laboratory has rigorous genetic quality control and mutant gene genotyping programs to ensure the genetic background of JAX® Mice strains as well as the genotypes of strains with identified molecular mutations. JAX® Mice strains are only made available to researchers after meeting our standards. However, the phenotype of each strain may not be fully characterized and/or captured in the strain data sheets. Therefore, we cannot guarantee a strain's phenotype will meet all expectations. To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
