These Esr2 knock-out mice exhibit female infertility. They are suitable for use in applications related to the study of the estrogen hormone signalling pathway.

Estrogen receptor 2 is one of two estrogen receptors, a ligand-activated transcription factor and nuclear hormone receptor. These mice carry a knock out mutation of the Esr2 gene in which exon 3 has been deleted. Homozygotes are viable, but female homozygotes are infertile, primarily due to reduced ovulation and defective ovarian function. Significantly reduced mRNA gene product is detected in ovaries of homozygotes. No protein gene product is detected by immunohistochemistry of ovaries from homozygotes.
A targeting vector containing a floxed TK-neo selection cassette was inserted downstream of exon 3 and a third loxP site was inserted upstream of exon 3. The construct was electroporated into an unspecified embryonic stem (ES) cell line (Xenogen) which were transiently transfected with a Cre recombinase vector to remove the selection cassette. ES cells that had successfully undergone Cre-mediated recombination and no longer retained the cassette or exon 3 were injected into blastocysts. The resulting chimeric animals were crossed to C57BL/6 mice, and then backcrossed to C57BL/6NCrl using a marker assisted protocol. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6NJ (Stock No. 0005304) at least once to establish the colony.

When maintaining a live colony, heterozygous mice may be bred together, to wildtype siblings, or to C57BL/6NJ inbred mice (Stock No. 005304). Homozygous females are infertile.
