Description

Egfl7:eGFP transgenic mice may be useful to track endothelial progenitors during early embryonic development and as a reporter of active angiogenesis in embryos and adults.

Detailed description

Egfl7:eGFP transgenic mice contain 5.4kb of the Epidermal growth factor-like domain 7 (Egfl7) promoter/enhancer sequences driving expression of an enhanced green fluorescent protein, EGFP. Hemizygotes are viable and fertile. Homozygous mice are not produced as a result of hemizygous intercrosses. Endogenous EGFL7 is expressed in the developing embryo and is largely restricted to sites of mesodermal progenitors of angioblasts/hemangioblasts and the vascular endothelium. Under control of Egfl7 regulatory sequences, EGFP labels sites of vasculogenesis and angiogenesis in the allantois, yolk sac, and in the embryo proper. Expression is also seen in adults in the uterus, ovary, and in neovasculature. EGFL7 is found to contribute to pathologic revascularization in the mouse retina. Expression can be monitored by direct visualization and immunofluorescence staining.

Development

The Egfl7:eGFP transgene was designed with an enhanced green fluorescent protein, EGFP, driven by 5.4kb of the Epidermal growth factor-like domain 7 (Egfl7) promoter/enhancer sequences, followed by an SV40 polyadenylation sequence. The transgene was microinjected into fertilized oocytes and mice from founder line 12 were bred to CD1 outbred mice to establish a colony. Upon arrival at The Jackson Laboratory, transgenic mice were bred to C57BL/6J inbred mice (Stock No. 000664) to establish the colony.
