These Uts2r knock-out mice exhibit abnormal vasoconstriction. They are suitable for use in applications related to the study of hypertension.

The Uts2r gene encodes for a G-protein coupled receptor for urotensin II. These mice carry a knock out mutation of the Uts2r gene in which the single coding exon is replaced by a PGK-NEO cassette. Mice that are homozygous for the targeted mutation are viable and fertile. Aortas isolated from homozygotes do not constrict when treated with human urotensis-II. On a high fat diet, homozygotes display decreased liver mass and hepatic steatosis, increased weight gain, and elevated serum triglyceride and LDL & HDL cholesterol content levels, when compared to wildtype controls.
When crossed to mice carrying a knock out mutation for the Apoe gene (see Stock No. 002052), the resulting double mutant mice exhibit more severe atherosclerosis, hyperlipidemia, and hyperinsulinemia, when compared to the Apoe knock out mice.
A targeting vector containing a PGK-NEO cassette was used to disrupt the single coding exon of the targeted gene. The construct was electroporated into 129P2/OlaHsd derived E14.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting male chimeric animals were bred to C57BL/6J females to achieve germline transmission. Heterozygotes were intercrossed to generate homozygotes.
Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
