Slc8a1 (solute carrier family 8 (sodium/calcium exchanger), member 1; also called Ncx1) is associated with Na+-Ca2+ exchange functionality. Animals that do not express the gene are embryonic lethal.
Exon 11 of the mouse Slc8a1 gene was flanked by loxP sites in this conditional mutant strain. Cre-mediated excision of the floxed region, which encodes part of the α-2 region essential for Na+-Ca2+ exchange activity, results in a knockout allele that leaves the reading frame intact.
When floxed mice are crossed with skeletal muscle-specific Myl1-cre mice (Stock No. 024713), and further bred to Sgcd knockout animals (Stock No. 004582), the severity of a muscular dystrophy phenotype is reduced.
Exon 11, encoding amino acids 722 to 813 and encompassing two transmembrane segments including part of the α-2 region essential for Na+-Ca2+ exchange activity, was flanked by loxP sites in an unspecified embryonic stem cell line. These mice were backcrossed to C57BL/6.
