This Trap1 knockout strain is useful in studies of cellular bioenergetics, mitochondrial homeostasis, tumor metabolism, and mechanisms of ageing.

The targeted Trap1 gene encodes a heat shock protein family mitochondrial chaperone protein that has a role in mitochondrial homeostasis, glycolysis regulation and tumor metabolism.
Homozygotes are viable and fertile. No gene product (protein) is detected by Western blot analysis of liver, kidney, uterus, spleen, brain, heart, and testis tissue from homozygotes.
Mice homozygous for the allele exhibit decreased levels of glucose and creatinine in blood, when compared to controls. Age related pathologies, such as hyperplasia, chronic inflammation, hepatic lipidosis, pancreatic fibrotic degeneration, and tumors observed in wildtype mice occur at significantly reduced incidence in homozygous mutant mice at 15-months of age.
The rsFROSAbgeo0s gene trap vector (incorporating fused beta galactosidase and neomycin, with inward-facing frt::f3::lox5171::loxP sites flanking the beta-gal-pA fusion protein) was introduced to 129P2/OlaHsd-derived E14TG2a.4 embryonic stem (ES) cells to generate mutant cell lines. Clone E150H04 ES cells (German Gene Trap Consortium), carrying a vector insertion downstream of the first exon of the Trap1 gene, were injected into blastocysts.
The resulting male chimeric animals were crossed to C57BL/6 female mice to generate heterozygotes. The mice were maintained on a mixed B6;129P2 background.
Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
