Description

These mice express Cre-ER^T2 under the direction of the murine Neurod1 promoter making them useful for spatial- and temporal-specific recombination in Neurod1-expressing cells during development or adulthood.

Detailed description

Mice homozygous for the BAC Neurod1-CreER^T2 transgene are viable and fertile. Expression of Cre-ER^T2 is directed by the murine (neurogenic differentiation 1) Neurod1 promoter/enhancer regions. NEUROD1 is a transcription factor involved in developmental programs of the gastrointestinal tract, pancreas, neurosensory, and central nervous system. Restricted to the cytoplasm, Cre-ER^T2 can only gain access to the nuclear compartment after exposure to tamoxifen. When these mice are bred with mice containing loxP-flanked sequence, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing cells of the offspring.

For example, when bred to mice expressing cre-inducible enhanced green fluorescent protein (EGFP), cre activity is seen in the resulting mice at E13.5 in neural tube, eye, olfactory epithelium, cranial ganglia, and dorsal root ganglia. In adults expression is seen in the neurogenic niche of the hippocampus and in a subset of mature neurons in the cortex, olfactory bulb, thalamus, hypothalamus, and cerebellum.

Development

Bacterial artificial chromosome (BAC) library (Source BioScience) was used to obtain a 160 kb BAC (BMQ-305H2) containing the entire mouse Neurod1 (neurogenic differentiation 1) gene. This BAC was modified to replace the coding region of Neurod1 with a creER^T2 fusion gene (a Cre recombinase fused to a human estrogen receptor ligand binding domain) followed by a frt-flanked neomycin (neo) selection cassette. The modified BAC was microinjected into fertilized C57BL/6JOlaHsd oocytes. Neurod1-CreER^T2 founder line M1, with intermediate levels of creER^T2 expression, was established and then subsequently maintained by breeding transgenic mice with C57BL/6JOlaHsd mice for at least 4 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
