Mice hemizygous for the PAC Gsh2-cre transgene are viable and fertile. These mice express codon optimized iCre directed by the endogenous Gsx2 (GS homeobox 2) promoter/enhancer regions on the PAC transgene. Cre recombinase expression is seen in precursor cells in the lateral, caudal, and medial ganglionic eminences as well as the septum. When crossed with a strain containing a loxP site-flanked sequence, iCre-mediated recombination results in deletion of the flanked sequence in subcortical precursors and all the neuronal and glial cell progeny.
A P1-derived artificial chromosome (PAC) containing the entire mouse Gsx2 (GS homeobox 2) gene was modified by targeted mutation to harbor codon optimized Cre Recombinase (iCre) sequence, followed by a polyadenylation sequence. A 110kb fragment was isolated from the PAC and was injected into C57BL6/CBA F1 donor oocytes. Resulting offspring were bred to C57BL6/CBA mice for at least 20 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J (Stock No. 000664) for at least one generation to establish the colony.

When maintaining a live colony hemizygous mice may be bred to wildtype (noncarrier) mice from the colony or to C57BL/6J (Stock No. 000664).

We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
