Myf6CE mice express a Cre-ERT2 fusion protein from endogenous Myf6 promoter/enhancer elements. MYF6 is a DNA binding protein involved in the development of muscle. Heterozygous mice are viable and fertile. When Myf6CE mice are bred with mice containing loxP-flanked sequence, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the floxed sequences.
For example, when bred to B6.129S4-Gt(ROSA)26Sortm1Sor/J mice (Stock No. 003474), tamoxifen-inducible cre-mediated recombination results in lacZ staining of somite from E8.75 to E10.75, and in trunk and limb muscles with increasing intensity from E12.75 to E14.75. lacZ staining was evident in the limb muscles of adult animals, and was absent from the heart.
A targeting vector was designed to replace exon 1e of the myogenic factor 6 (Myf6) gene with a CreERT2 (Cre recombinase fused to an estrogen receptor ligand binding domain) coding sequence and a frt-flanked neomycin (neo) resistance cassette. This construct was electroporated into C57BL/6;129sv embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to Tg(ACTFLPe)9205Dym/J mice to remove the neo selection cassette. Offspring were then crossed to remove the Flp-expressing transgene. These mice were bred to 129svJ mice for at least 3 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

When maintaining a live colony heterozygous mice may be bred to wildtype mice from the colony. The donating investigator has not attempted to make this strain homozygous.
