FN2 mice have exons 3-5 of the neuroligin 2 (Nlgn2) gene floxed, and may be useful for studying synapse formation and/or function.

These FN2 floxed mice possess loxP sites flanking exons 3-5 of the neuroligin 2 (Nlgn2) gene. Mice that are homozygous for this allele are viable and fertile. NLGN2 is a transmembrane scaffolding protein involved in cell-cell interactions, and plays a role in synapse function and synaptic signal transmission. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 3-5 deleted in the cre-expressing tissues. When expression of cre in the prefrontal cortex was induced by local injections of viral vectors, the NLGN2 protein was reduced to undetectable level at the infected region.
A targeting vector was designed to insert a loxP site followed by a frt-flanked neomycin resistance (neo) cassette upstream of exon 3, and a second loxP site downstream of exon 5 of the neuroligin 2 (Nlgn2) gene. The construct was electroporated into C57BL/6NTac-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J transgenic mice (Stock No. 003800) to delete the neo cassette. Progeny were crossed to remove the Flp-expressing transgene and resulting FN2 floxed mice were bred together for at least 4 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
