Detailed description

Med23 (mediator complex subunit 23) is a subunit of the Mediator coactivator complex, and is important in the expression of a subset of MAPK/ERK pathway-responsive genes.

Exons 5-7 of the mouse Med23 gene are flanked by loxP sites in this conditional targeted mutant strain. Cre excision of the floxed region results in a knockout of the gene.

Thymocyte-specific knockouts of the gene created through crosses with Lck-cre produce T cells that fail to efficiently populate peripheral lymphoid organs.

Development

An FRT-flanked neomycin cassette was placed in intron 4 and exons 5-7 of the gene were flanked by loxP sites through homologous recombination in 129S6/SvEvTac-derived TL-1 embryonic stem (ES) cells. Transient expression of FLP recombinase in the ES cells excised the neomycin cassette, leaving floxed exons 5-7. Resultant chimeric mice were crossed with C57BL/6J animals, then to 129P2/Ola for 3 generations. Fertility/productivity issues precipitated 18 generations of backcrossing to 129S2/SvPasCrl by the donating laboratory.
