Lats1f/f floxed mice possess loxP sites flanking exon 4 of the large tumor suppressor (Lats1) gene. LATS1 is a serine/threonine-protein kinase involved in the regulation of the Hippo pathway. Hippo signaling pathway plays a role in organ size control and tumor suppression by restricting proliferation and promoting apoptosis. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 4 deleted in the cre-expressing tissues.
For example when these mice are bred to mice carrying a floxed-Lats2 allele (Stock No. 025428) and afterwards bred to B6.FVB(129)-Tg(Myh6-cre/Esr1*)1Jmk/J mice (Stock No. 005657), tamoxifen-inducible Cre-expression results in Lats 1/2 deficient cardiomyocytes which exhibit enhanced heart regeneration and cardiomyocyte renewal.
A targeting vector was designed to flank exon 4 of the large tumor suppressor (Lats1) gene with loxP sites. This construct also contained a frt-flanked neomycin resistance (neo) cassette. The construct was electroporated into (C57BL/6-Tyrc-Brd x 129S6/SvEvTac)F1-derived RJ2.2 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to CD-1 outbred mice. Offspring were bred with Flp transgenic mice to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene, resulting in a colony homozygous for the Lats1f/f allele. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
