These tetO-EGFP-L10a mice carry a EGFP/Rpl10a fusion gene, regulated by a tetracycline operator (tetO). When crossed with a mutant strain expressing tetracycline-controlled transactivator protein (tTA), this strain allows identification and collection of specific changes in local mRNA populations by ribosome immunoprecipitation.

Expression of the epitope-tagged EGFP/Rpl10a fusion gene (EGFP fused to the N-terminus of the large subunit ribosomal protein L10a) is regulated by a tetracycline operator (tetO) in this transgenic strain. When mated to a strain expressing tetracycline-controlled transactivator protein (tTA), expression of EGFP/Rpl10a fusion protein may be regulated with tetracycline or its analog doxycycline (dox) in the double mutant offspring. Expression of EGFP/Rpl10a within a cell type allows identification and collection of cell type-specific changes in mRNA populations by ribosome immunoprecipitation, where recovery of intact cells for analysis is not possible.
Mice that are hemizygous for the transgene are viable and fertile.
When crossed with a Camk2a-tTA strain that expresses tTA in forebrain neurons (see Stock No. 007004, for example), ribosome-bound mRNA from CA1 pyramidal neuron dendrites can be isolated and collected by immunoprecipitating EGFP-tagged ribosomes.
A transgenic construct containing sequence encoding EGFP/Rpl10a fusion protein (EGFP fused to the N-terminus of the large subunit ribosomal protein L10a) under the control of the tetO, tetracycline-responsive regulatory element, was injected into fertilized C57BL/6J mouse eggs. Founder line 5a was subsequently established. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

When maintaining a live colony, hemizygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). The Donating Investigator has not attempted to make the strain homozygous.

We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
