Aquaporin 11 (Aqp11 ) encodes a membrane channel of undetermined function that colocalizes with markers of the rough endoplasmic reticulum (RER). These mice possess loxP sites flanking exons 2 and 3. Mice that are homozygous for this allele are viable and fertile. When crossed to mice carrying Tg(Alb-cre)21Mgn, Cre recombinase expression in the liver has little effect beyond a minor defect in lipid handling in unchallenged mice. However, challenge by fasting/re-feeding or administration of high dose free amino acids results in RER-derived vacuoles in periportal hepatocytes.
Complete deficiency of Aqp11 causes death by 2-8 weeks of age due to kidney malfunction.
This conditional knockout may be useful for studying the role of aquaporin 11 in liver and other organs.
The targeting vector contains a loxP site upstream of exon 2 and a loxP site downstream of exon 3 followed by a frt-flanked PGKneo cassette. The construct was electroporated into 129S1/Sv-derived CJ7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. Transient infection with a FLPe plasmid excised the neomycin cassette. The resulting chimeric animals were crossed to C57BL/6 mice. The strain is maintained on a mixed background. Upon arrival, mice were bred to C57BL/6J (Stock No. 000664) mice for at least 1 generation to establish the colony.

While maintaining a live colony, these mice are bred as homozygotes.

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
