Description

This loxP-flanked aquaporin 11 (Aqp11) strain may be useful for studying the role of aquaporin 11 in liver and other organs.

Detailed description

Aquaporin 11 (Aqp11 ) encodes a membrane channel of undetermined function that colocalizes with markers of the rough endoplasmic reticulum (RER). These mice possess loxP sites flanking exons 2 and 3. Mice that are homozygous for this allele are viable and fertile. When crossed to mice carrying Tg(Alb-cre)21Mgn, Cre recombinase expression in the liver has little effect beyond a minor defect in lipid handling in unchallenged mice. However, challenge by fasting/re-feeding or administration of high dose free amino acids results in RER-derived vacuoles in periportal hepatocytes. Complete deficiency of Aqp11 causes death by 2-8 weeks of age due to kidney malfunction. This conditional knockout may be useful for studying the role of aquaporin 11 in liver and other organs.

Development

The targeting vector contains a loxP site upstream of exon 2 and a loxP site downstream of exon 3 followed by a frt-flanked PGKneo cassette. The construct was electroporated into 129S1/Sv-derived CJ7 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. Transient infection with a FLPe plasmid excised the neomycin cassette. The resulting chimeric animals were crossed to C57BL/6 mice. The strain is maintained on a mixed background. Upon arrival, mice were bred to C57BL/6J (Stock No. 000664) mice for at least 1 generation to establish the colony.
