Expression of the calcium indicator, GCaMP6s, is regulated by the tetracycline-responsive regulatory element (tetO) in these mice. When mated with a strain expressing tetracycline-controlled transactivator protein (tTA), expression of the GCaMP6s protein may be regulated with the tetracycline analog doxycycline (dox) in the double-allelic offspring. This strain may be useful in detecting single neuronal action potentials.

These tetO-GCaMP6s transgenic mice express the genetically encoded calcium indicator, GCaMP6s, under the control of a tetracycline-responsive promoter element (TRE; tetO). When hemizygotes are bred with another transgenic mouse expressing either reverse tetracycline-controlled transactivator protein (rtTA) or tetracycline-controlled transactivator protein (tTA) GCaMP6s expression can be regulated with the tetracycline analog, doxycycline, with GCaMP6s expression approximately, though not completely, recapitulating the expression pattern of the tTA driver. The GCaMP6s indicator is an ultrasensitive detector of single neuronal action potentials and has slow response kinetics. GCaMP consists of a circularly permutated green fluorescent protein (cpGFP), the calcium-binding protein calmodulin (CaM), and CaM-interacting M13 peptide. Hemizygous transgenic mice are viable and fertile. The Donating Investigator reports that homozygotes are viable and fertile (July 2017).
A transgenic construct containing the genetically encoded calcium indicator, GCaMP6, under the control of the tetO, tetracycline-responsive regulatory element, was injected into fertilized B6;DBA mouse eggs. Founder line G6s2 was subsequently established. The mice were then backcrossed to C57BL/6 for 1 generation. The Donating Investigator reports that the transgene inserted into Chromosome 13, between 5710045 and 5710046. During backcrossing, the Y chromosome was fixed to the C57BL/6 genetic background. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

When maintaining a live colony, hemizygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). The Donating Investigator reports that homozygotes are viable and fertile (July 2017).
