COX10Flx mutant mice possess loxP sites flanking exon 6 of the Cox10 gene. This strain may be useful for generating conditional mutations in applications related to the study of mitochondrial myopathy and mitochondrial respiratory chain deficiency.

The Cox10 gene encodes for the cytochrome c oxidase (COX) assembly protein mitochondrial heme A:farnesyltransferase and is essential for COX functioning.
These mice possess loxP sites on either side of exon 6 of the targeted Cox10 gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 6 deleted in the cre-expressing tissues.
When bred to a strain with Cre recombinase expression in skeletal muscle (See Stock No. 024713, for example), this mutant mouse strain may be useful in studies of progressive mitochondrial myopathy.
When bred to a strain with Cre recombinase expression in the forebrain and cerebellum of the embryo, this mutant mouse strain may be useful in studies of mitochondrial encephalopathy.
A loxP site flanked targeting vector containing PGK-Neo selection cassette was utilized in the construction of this mutant. This selection cassette was inserted upstream of exon 6 of the targeted gene, and another loxP site was inserted downstream of exon 6. This construct was electroporated into 129X1/SvJ derived GSI-1 embryonic stem (ES) cells which were transiently transfected with a Cre recombinase vector to remove the selection cassette. ES cells that had successfully undergone Cre mediated recombination and no longer retained the cassette but did retain the loxP-flanked exon 6 were injected in C57BL/6 blastocysts. Resulting chimeric male animals were backcrossed to wildtype C57BL/6 mice. The mice were then backcrossed to C57BL/6J for 10 generations.
Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
