These transgenic mice express a tamoxifen-inducible cre recombinase from a Adipoq promoter. They may be useful in generating adipocyte-specific targeted mutants in studies related to adipose tissue function and storage, obesity, and other metabolic diseases.

Mice hemizygous for this Adipoq-CreER BAC transgene are viable and fertile, with expression of a tamoxifen inducible Cre recombinase (cre/ERT) directed to adipose tissue by the promoter/regulatory regions of the mouse adiponectin (Adipoq) locus on the BAC transgene. Transcription/translation from the BAC Adipoq locus is disabled. Expression levels of cre/ERT are similar to that of endogenous Adipoq expression. Over 80% of adipocytes in all adipose depots exhibit Cre recombinase activity after tamoxifen injection. Transgene expression may also occur in bone, although this tissue has not yet been tested. Cre recombinase activity is not detected in the absence of tamoxifen.
To generate the Adipoq-CreER BAC transgenic mice, the 245 kb RP23-90G21 bacterial artificial chromosome (BAC) containing at least five mouse genes was modified by replacing the starting ATG and 222 bp of the adiponectin gene with the starting ATG and sequence encoding the cre/ERT, Cre recombinase and estrogen receptor 1 (mouse) fusion gene. This modified BAC transgene was microinjected into fertilized 129 mouse eggs. Founder line 1 was subsequently established. The mice were then backcrossed to C57BL/6J wildtype mice for more than 15 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

When maintaining a live colony, hemizygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). The Donating Investigator has not attempted to make the strain homozygous.
