This allele creates tricistronic expression of the endogenous OR8A1 (formerly called OLFR151), a constitutively active GNAS, and a GFP fluorescent marker all from the Or8a1 locus. This permits assessment of the impact of increased GNAS activity on the maturation and function of olfactory sensory neurons expressing wild-type OR8A1. Histology of homozygotes found 16 of 36 olfactory bulbs had a GFP+ lateral glomerulus in the correct location for an OR8A1 glomerulus, but in 20 of those 36 there was failure of the GFP+ axons to converse into a glomerulus. Additionally, compound heterozygotes between this allele and Or8a1tm27Mom, which has tauRFP2 bicistronically expressed with Or8a1, show axon migration to the same location, but these form compartmentalized glomeruli instead of unifying into a single glomerulus, indicative of distinct identities.
