These floxed mutant mice possess loxP sites flanking exon 4 of the Rest gene. This strain may be useful for generating conditional mutations in applications related to the study of neurogenesis.

These REST/NRSFloxP mice possess loxP sites on either side of exon 4 of the targeted gene. The Rest gene encodes for a transcriptional repressor that has an important role in neurogenesis. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 4 deleted in the cre-expressing tissues.
When bred to a strain with inducible Cre recombinase expression the adult and developing mouse brain (see Stock No. 016261), this mutant mouse strain may be useful in studies of adult neuroregeneration.
A targeting vector containing a FRT site flanked PGK-Neo selection cassette and a 3' loxP site was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 4 of the targeted gene, and another loxP site was inserted upstream of exon 4. This construct was electroporated into 129S6/SvEvTac derived SM1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6N blastocysts. The resulting chimeric animals were crossed to hACTB:FLPe transgenic mice (on a 129SV/J genetic background) expressing FLP recombinase to remove the FRT flanked selection cassette. Mice that retained the loxP site flanked exon 4 were then bred to C57BL/6J mice for at least 10 generations. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.

When maintaining a live colony, these mice can be bred as homozygotes.
