These floxed mutant mice possess loxP sites flanking exon 2 of the Nlk gene. This strain may be useful for generating conditional mutations in applications related to the regulation of bone development, and Wnt/beta-catenin, BMP/Smad and Notch signaling.

These mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues. Exon 2 encodes for part of the kinase domain with excision of exon 2 resulting in a frameshift. The serine/threonine-protein kinase produced by the Nlk gene is related to mitogen-activated protein kinases and can regulate Wnt/beta-catenin, BMP/Smad and Notch signaling.
When bred to a strain with Cre recombinase expression in osteoblast precursor cells (see Stock No. 005657 for example), this mutant mouse strain may be useful in studies of bone homeostasis.
A targeting vector containing a FRT site-flanked PGK-Neo selection cassette and a downstream loxP site was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into 129SvJ/C57BL/6 derived F1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts.
The resulting chimeric male animals were crossed to FLP recombinase expressing mice (Stock No. 003946) to remove the FRT-flanked selection cassette. The mice were then crossed to wildtype C57BL/6J mice to breed away from the FLP recombinase allele. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
