These mice possess loxP sites on either side of exon 41 of the Lrrk2 gene. This strain may be useful for generating conditional mutations suitable for use in applications related to the study of dendritic neuronal arborization and Parkinson's disease.

Mutations in the human LRRK2 gene have been found to cause autosomal dominant Parkinson disease-8. These mice possess loxP sites on either side of exon 41 of the Lrrk2 (leucine-rich repeat kinase 2) gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 41 deleted in the cre-expressing tissues.
A targeting vector containing a FRT flanked PGK-NEO cassette and a loxP site was inserted downstream of exon 41. A second loxP site was inserted upstream of exon 41. The construct was electroporated into C57BL/6 derived Bruce4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into BALB/cJ blastocysts. The resulting chimeric animals were crossed to C57BL/6J mice.
Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony. The mice were then crossed to FLP deleter mice Stock No. 005703) to remove the FRT site flanked NEO cassette. Further breeding to C57BL/6J (Stock No. 000664) was done to remove the FLP recombinase allele.

When maintaining a live colony, these mice can be bred as homozygotes.
