Description

These RHAU^fl/fl mice may be useful for studying the role of DXH36 helicase in the differentiation of hematopoietic cells lineages.

Detailed description

These RHAU^fl/fl mutant mice possess loxP sites flanking exon 8 of the DEAH (Asp-Glu-Ala-His) box polypeptide 36 (Dhx36) gene. RHUA is an RNA helicase that plays a role in the deadenylation and decay of mRNAs with 3'-UTR AU-rich elements. RHUA is critical for mouse hematopoiesis. Mice that are homozygous for this allele are viable and fertile. Breeding these mice to mice that express germ line Cre results in embryonic lethality. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 8 deleted in the cre-expressing tissues.

For example, breeding to B6.Cg-Tg(Vav1-cre)A2Kio/J mice (Stock No. 008610) results in a truncated RHAU protein and leads to hemolytic anemia and differentiation defect at the proerythroblast stage.

Development

A targeting vector was designed to insert a loxP site upstream of exon 8 of the DEAH (Asp-Glu-Ala-His) box polypeptide 36 (Dhx36) gene, followed by a frt-flanked neomycin resistance (neo) cassette, and a second loxP site downstream of exon 8. The construct was electroporated into 129P2/Ola-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to C57BL/6 mice. Offspring were bred with 129S4/SvJaeSor-Gt(ROSA)26Sor^tm1(FLP1)Dym/J (Stock No. 003946) transgenic mice to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. The donating investigator reported that these mice were bred to BALB/cJ mice for at least 5 generations (see SNP note below). Upon arrival, mice were bred to BALB/cJ inbred mice (Stock No. 000651) for at least one generation to establish the colony.

A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Fifteen of the 27 markers were segregating with 129 or B6, suggesting an incomplete backcross.
