Description

These ATGL-flox mutant mice possess loxP sites flanking exon 2 through 7 of the Pnpla2 gene. This strain may be useful for generating conditional mutations in applications related to the study of the regulation of lipid and energy homeostasis.

Detailed description

The Pnpla2 gene encodes for the adipose triglyceride lipase enzyme that catalyzes the first step in triglyceride hydrolysis. These mice possess loxP sites on either side of exons 2 through 7 of the Pnpla2 gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 2 through 7 deleted in the cre-expressing tissues, resulting in loss or significant reduction of ATGL-mediated triacyglycerol hydrolysis in the tissues where Cre recombinase is expressed.

Successful tissue-specific Cre-mediated recombination of the floxed ATGL allele in this strain has been verified in cardiac and skeletal muscle (see Stock No. 006475 for example), adipose tissue (see Stock No. 010803), liver (see Stock No. 003574) and intestinal epithelial cells (see Stock No. 004586).

Development

A FRT site flanked targeting vector containing PGK-Neo selection cassette was utilized in the construction of this mutant. This selection cassette and a loxP site was inserted downstream of exon 7 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into unspecfied 129S derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to mice (on the 129S4/SvJaeSor genetic background) expressing FLP1 recombinase (Stock No. 003946). Mice that retained the loxP site flanked exons 2 through 7 were then backcrossed to C57BL/6NTac mice for more than 10 generations. Heterozygotes were crossed to generate homozygotes. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6NJ (Stock No. 005304) at least once to establish the colony.
