These BAC transgenic mice carry a single copy of Flag-tagged Hist2h2be (histone cluster 2, H2be), likely integrated on the X-chromosome. Flag-H2be mice have no apparent phenotype differences from wildtype littermates, but males do not efficiently transfer the transgene. Embryonic viability may be compromised among female embryos carrying the transgene contributed from the father.

Hist2h2be (histone cluster 2, H2be) is expressed exclusively in olfactory chemosensory neurons. Expression is reduced by sensory activity, and it promotes neuronal cell death, potentially modulating olfactory neuron population dynamics to adapt the olfactory repertoire to the environment.
These BAC transgenic mice carry a single copy of Flag-tagged Hist2h2be, likely integrated on the X-chromosome. Expression of the Flag-H2be transgene was examined by RT-qPCR in the main olfactory epithelium (MOE), where the expression level of the transgenic allele was found to be identical to that of the endogenous Hist2h2be allele. Expression was also examined by immunofluorescence and in situ hybridization in the MOE, vomeronasal organ, and brain, revealing indistinguishable patterns of expression (or lack of expression, in the case of the brain) for FLAG-H2BE protein and endogenous Hist2h2be mRNA. Males do not efficiently transfer the transgene (see maintenance note for more details).
A Flag-encoding DNA sequence was inserted upstream of the Hist2h2be coding region within BAC RP23-16G3, which contains a 200-kb region of mouse genomic sequence surrounding the gene. The modified BAC was amplified in E. coli, confirmed by sequencing, and injected into B6D2F1 (C57BL/6J x DBA/2J) embryos. Transgenic founders were crossed to C57BL/6J mice for approximately 5 generations to establish founder line 2 which carries 1 copy of the transgene.

Hemizygotes are viable and fertile. The transgene is reportedly transferred from the father very inefficiently. The transgene is likely located on the X-chromosome, as males produced from a cross between a hemizygous male and a wild type female are invariably wild type. In addition, very few females are produced from the same cross, indicating that there may be a viability problem among female embryos carrying the transgene from the father (although there is no problem transferring the transgene from the mother). These issues may make in vitro fertilization from cryopreserved transgenic sperm problematic.
