This transgenic strain enables conditional cell-specific expression of tetanus toxin light chain under the control of a tetracycline sensitive (tetO) promoter.

These mice express tetanus toxin light chain under the control of a tetracycline sensitive (tetO) promoter. The tetanus toxin light chain is fused to an internal ribosome entry site (IRES), and a Tau-β-galactosidase (lacZ) fusion protein. Tetanus toxin light chain is a molecule that cleaves synaptic vesicle protein VAMP2 inhibiting synaptic release. Neurons directed to express the toxin through crosses with neuron-specific tetracycline-expressing (tTA) strains fail to maintain appropriate synaptic connections and are eliminated through competition with active neurons. Conditional lacZ staining is evident in inactivated neurons. Hemizygous mice are viable and fertile.
When bred to a strain expressing tTA in the neurons of the olfactory bulb (Omptm1(tTA)Gogo - see Stock No. 017754 for example), mice treated with doxycycline exhibit inhibition of synaptic release in olfactory sensory neurons.
The TeTxLC-IRES-tau-lacZ transgene was designed with the tetracycline operator (tetO or tet-operator) driving expression of a tetanus toxin light chain fused to an internal ribosome entry site (IRES), and a Tau-β-galactosidase (lacZ) fusion protein. This transgene was microinjected into oocytes and founder mice were bred to C57BL/6 mice to establish the colony. Upon arrival at The Jackson Laboratory, transgenic mice were bred with C57BL/6J inbred mice (Stock No. 000664) for at least one generation.
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. The 27 markers throughout the genome suggested a C57BL/6 genetic background and all 5 markers that determine C57BL/6J from C57BL/6N were found to be C57BL/6J. These data suggest the mice sent to The Jackson Laboratory Repository were on a congenic C57BL/6 genetic background.
