These Fgf10c mice possess loxP sites flanking exon 2 of the fibroblast growth factor 10 (Fgf10) gene. The donating investigator reports that a tetracycline response element present upstream of exon 2 has no effect on expression unless a tetO binding protein is present and activated. FGF10 is expressed in head and limb mesenchyme, and is involved in cell division, embryonic development, regulation of cell growth and maturation, formation of blood vessels, and wound healing. Homozygotes are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues.
For example, when bred to BALB/c-Tg(CMV-cre)1Cgn/J mice (Stock No. 003465), FGF10 null mice fail to initiate inner ear and limb development.
A targeting vector was designed to insert a tetracycline response element, a single loxP site, and a frt-flanked neomycin (neo) and kanamycin (kan) resistance cassette upstream of exon 2 of the fibroblast growth factor 10 (Fgf10) gene. A second loxP site was inserted downstream of exon 2. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric males were bred to C57BL/6 mice. Offspring were bred with
129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym/J transgenic mice (Stock No. 003946) to delete the selection cassette, and progeny were crossed to remove the Flp-expressing transgene. These mice were maintained on a mixed background. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
