Detailed description

These floxed-AP-2gamma mutant mice possess loxP sites flanking exon 6 of the transcription factor AP-2, gamma (Tfap2c) gene. AP-2gamma is a transcription factor expressed early in embryogenesis in trophoblast cell lineage. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 6 deleted in the cre-expressing tissues.

Development

A targeting vector was designed to insert a loxP site upstream of exon 6 of the transcription factor AP-2, gamma (Tfap2c) gene. A frt-flanked neomycin resistance (neo) cassette, followed by a second loxP site were placed downstream of exon 6. The construct was electroporated into 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺-derived CJ-7 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a actb-Flpe expression plasmid to delete the neo cassette. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric mice were bred to Black Swiss mice. These mice were subsequently crossed to mice carrying the Tfap2a^tm2Will allele on a mixed Swiss Black x 129S background. Upon arrival, mice were bred to 129S1/SvImJ inbred mice (Stock No. 002448) for at least one generation to establish the colony. The Tfap2a^tm2Will allele was bred out of this colony, and is maintained as Stock No. 023406.
