Description

These floxed mutant mice possess loxP sites flanking exon 2 of the Pik3r4 gene. This strain may be useful for generating conditional mutations in applications related to the regulation of endosomal sorting and autophagy.

Detailed description

These Vps15^f/f mutant mice possess loxP sites flanking exon 2 of the phosphatidylinositol 3 kinase, regulatory subunit, polypeptide 4, p150 (Pik3r4) gene. Vps15 (Vacuolar Protein Sorting 15) is involved in endosomal sorting and autophagy, which is required for tissue homeostasis and renewal. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues.

For example, when bred to B6.Cg-Tg(ACTA1-cre)79Jme/J (see Stock No. 006149) expressing Cre recombinase in skeletal muscle, resulting offspring display muscle weakness, the accumulation of autophagosomes, and glycogen and sarcolemmal features within the muscle fibers characteristic of autophagic vacuolar myopathies (AVM).

Development

A targeting vector was designed to insert a loxP site upstream of exon 2, and a second loxP site followed by a frt-flanked neomycin resistance (neo) cassette, downstream of exon 2 of the phosphatidylinositol 3 kinase, regulatory subunit, polypeptide 4, p150 (Pik3r4) gene. The construct was electroporated into 129S2/SvPas-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6NCrl blastocysts and resulting chimeric mice were obtained. Offspring were bred with Flp transgenic mice to delete the neo cassette, and progeny were bred to C57BL/6NCrl mice to remove the Flp-expressing transgene. These Vps15^f/f were bred to C57BL/6NCrl mice for at least 12 generations. Upon arrival, mice were bred to C57BL/6NJ inbred mice (Stock No. 005304) for at least one generation to establish the colony.
