Hemizygous Phox2b::FLPo BAC transgenic mice are viable and fertile, with FLP recombinase expression under control of the Phox2b promoter/enhancer regions within the BAC transgene. Flp-expressing neurons co-express Phox2B (as shown by reported gene expression). Phox2b-FLPo BAC transgenic mice from founder line 3276 exhibit FLP expression directed to central and peripheral neurons that control cardiovascular, digestive and respiratory systems. When bred with mice containing a frt-flanked sequence of interest, FLP-mediated recombination will result in deletion of the flanked sequences in the offspring. These Phox2b::FLPo BAC transgenic mice may be used to generate conditional mutations for studying neuronal cell types depending on Phox2b that control feeding, glucose homeostasis, blood pressure and autonomic respiration. The Phox2b::FLPo BAC transgenic line, when paired with a suitable Cre driver and a dual Cre- and FLP-responsive reporter allele will be useful for intersectional genetic fate mapping of subpopulations of Phox2b-expressing neurons.
The C57BL/6J mouse bacterial artificial chromosome (BAC) RP24-95M11, containing the entire paired-like homeobox 2b Phox2b locus was obtained. This BAC also contained ~150 kbp sequence upstream of the Phox2b locus and ~50 kbp of sequence downstream of the Phox2b locus. A codon-optimized FLPo recombinase sequence fused with an SV40 polyadenylation signal, and a kanamycin resistance cassette were inserted into exon 2 of Phox2b. The resulting Phox2b::FLPo BAC transgene was injected into (C57BL/6 X DBA/2) F1 fertilized oocytes. Mice from founder line 3276 were subsequently backcrossed to (C57BL/6 X DBA/2) F1 mice. Upon arrival at The Jackson Laboratory Repository, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.

When maintaining a live colony, hemizygous mice may be bred to wildtype (noncarrier) mice from the colony, or to C57BL/6J inbred mice (Stock No. 000664).

To ensure that JAX® Mice will meet the needs of individual research projects or when requesting a strain that is new to your research, we suggest ordering and performing tests on a small number of mice to determine suitability for your particular project. We do not guarantee breeding performance and therefore suggest that investigators order more than one breeding pair to avoid delays in their research.
