ERM- mice may be useful for studying Etv5 transcriptional control during cell growth and differentiation, and stem cell self-renewal.

Exons 2-5 of the ets variant gene 5 (Etv5) gene have been removed in these ERM- mice, abolishing gene function. ERM is a transcription factor expressed in brain, ovaries, kidneys, skeletal muscle, lung, and Sertoli cells in the testis. ERM is required for cell growth and differentiation. Heterozygotes are viable and fertile. Homozygotes are viable but infertile, and exhibit a decrease in body weight and a progressive decrease in testis weight. These ERM- mice exhibit a loss of spermatogonial stem cell self-renewal and a failure to maintain spermatogonia, resulting in a Sertoli cell-only phenotype by 10 weeks of age. They develop muscle weakness and show functional neuromuscular transmission defects. When bred with mice lacking Etv4 expression, double mutant mice are completely absence of kidney development.
A targeting vector was designed to replace exons 2-5 of the ets variant gene 5 (Etv5) gene with a loxP-flanked neomycin resistance (neo) cassette. The construct was electroporated into 129S6/SvEvTac-derived MC50 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a cre expression plasmid to delete the neo cassette. Resulting ES cells were injected into blastocysts and resulting chimeric males were bred with 129SvEv females to establish a colony of ERM- mice (See SNP note below). Upon arrival, mice were bred to 129S1/SvImJ inbred mice (Stock No. 002448) for at least one generation to establish the colony.
A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Markers throughout the genome were segregating from an unknown source.
