These Tpp1flox mice possess loxP sites flanking exons 3-8 of the adrenocortical dysplasia (Acd) gene. Acd encodes a protein that is part of the telosome/shelterin telomeric complex which is involved in telomere maintenance and protection. Mice that are heterozygous for this allele are viable and fertile. The fertility of homozygotes has not been tested, but they are smaller and are not as vigorous as heterozygotes. When these mutant mice are bred to mice that express Cre, homozygous embryos die by E8.5 due to telomere dysfunction.
A targeting vector was designed to insert a loxP site upstream of exon 3 and a second loxP site followed by a frt-flanked neomycin resistance (neo) cassette, downstream of exon 8 of the adrenocortical dysplasia (Acd) gene. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCr)F1-derived G4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into B6(Cg)-Tyrc-2J/J blastocysts and resulting chimeric males were bred to B6(Cg)-Tyrc-2J/J females (Stock No. 000058). Offspring were bred with
B6.Cg-Tg(ACTFLPe)9205Dym/J (Stock No. 005703) mice to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. Resulting Tpp1flox mice were bred to C57BL/6 mice for at least 5 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation. The Tyrc-2J allele was bred out of this colony.

When maintaining a live colony, heterozygous mice may be bred to wildtype mice from the colony or to C57BL/6J inbred mice (Stock No. 000664). The fertility of homozygotes has not been tested, they are smaller and are not as vigorous as heterozygotes.
