These TAp63fl mice possess loxP sites flanking exon 2 of the transformation related protein 63 (Trp63) gene and have applications in studies related to epidermal development and differentiation, longevity, ageing, obesity, metabolism and cancer.

Trp63 (transformation related protein 63) encodes multiple isoforms, which have roles in longevity, ageing, obesity, metabolism, and regulation of epidermal development and differentiation. These mice possess loxP sites on either side of exon 2 of the targeted Trp63 gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues.
When bred to a strain with Cre recombinase expression in cells of the epidermal basal layer, the resulting mice prematurely age, and exhibit a phenotype including blisters, skin ulceration, and senescence of hair follicle associated dermal/epidermal cells.
Germline cre mediated deletion of TRP63 isoforms results in knock out mice that also are tumor prone and develop metastatic lesions.
A targeting vector designed by Dr. Elsa R. Flores (University of Texas M.D. Anderson Cancer Center) containing a FRT site flanked NEO cassette with a downstream loxP site was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 2. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCr)F1 derived G4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric male animals were crossed to C57BL/6 albino female mice.
Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
