These Rab7flox mutant mice possess loxP sites flanking exon 1 of the RAB7, member RAS oncogene family (Rab7) gene. RAB7 is a small GTPase that promotes fusion reactions between late endosomes and lysosomes facilitating the lysosomal degradation of plasma membrane proteins, autophagosomes, and endocytic vesicle cargo. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 1 deleted in cre-expressing tissues.
For example, when crossed to a strain expressing Cre recombinase in CD4 T cells (see Stock No. 017336), this mutant mouse strain may be useful for studying the effect of autophagosomes degradation on T cell survival.
A targeting vector was designed to insert a single loxP site upstream of exon 1, and a loxP-flanked neomycin resistance (neo) cassette downstream of exon 1 of the RAB7, member RAS oncogene family (Rab7) gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric males were bred with C57BL/6J females. Offspring were bred to a Protamine-Cre transgenic strain to delete the neo cassette. Resulting mice contained multiple gene rearrangments; intact floxed-exon 1, intact floxed-neo cassette, or excision of both exon 1 and the neo cassette. Mice, containing only the floxed-exon 1, were bred to C57BL/6J mice for at least 10 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
