BXSB.B6-Yaa+ consomic males carry the C57BL/6-derived Y chromosome in place of the mutant Yaa-containing Y chromosome of BXSB/MpJ inbred male mice. BXSB.B6-Yaa+ consomic mice are available with different amounts of backcrossing to BXSB/MpJ inbred females: Stock No. 021330 are backcrossed at least 40 generations, Stock No. 001925 are backcrossed at least 22 generations, and Stock No. 000742 are backcrossed at least 10 generations.
These BXSB.B6-Yaa+ mice are a BXSB-consomic strain that carries the C57BL/6-derived Y chromosome in place of the mutant Yaa-containing Y chromosome of BXSB/MpJ inbred male mice (Stock No. 000740). Male as well as female BXSB.B6-Yaa+ mice do not develop the severe systemic lupus erythematosus (SLE)-like autoimmune disease that spontaneously develops in BXSB/MpJ male mice. BXSB.B6-Yaa+ male mice are a sex-matched, autoimmune disease-resistant control strain for BXSB/MpJ male mice.
The Y-linked autoimmune accelerator locus (Yaa) on the Y chromosome of BXSB inbred male mice is the result of a duplication of an ~4 Mbp telomeric segment near the pseudoautosomal region of the X chromosome onto the Y chromosome. The duplicated segment contains 19 genes, including toll-like receptor 7 (Tlr7) and phosphoribosyl pyrophosphate synthetase 2 (Prps2). The BXSB-derived Yaa locus is attributed to the systemic lupus erythematosus (SLE)-like autoimmune disease characteristics observed in BXSB/MpJ inbred male mice.
These BXSB.B6-Yaa+ mice (Stock No. 021330) are a BXSB-consomic control strain that was bred to harbor the C57BL/6-derived wildtype allele of Yaa in place of the BXSB/MpJ-derived Yaa allele. The strain history is shown below.
First, BXSB/MpJ inbred females (Stock No. 000740) were bred to C57BL/6J inbred males (Stock No. 000664). Each successive generation, male offspring with the C57BL/6J-derived Y chromosome were bred with BXSB/MpJ inbred females. The congenic strain after ten generations of backcrossing is BXSB.B6-Yaa+/J (N10; Stock No. 000742).
Around 1998, Drs. Larry E. Mobraaten and Derry C. Roopenian (The Jackson Laboratory) obtained some BXSB.B6-Yaa+/J males from The Jackson Laboratory Repository and continued the backcrossing for additional generations. The congenic strain after at least 22 generations of backcrossing is BXSB.B6-Yaa+/MobJDcr (N22; Stock No. 001925).
In 2013, Dr. Roopenian sent some mice at generation N40 to The Jackson Laboratory Repository to establish Stock No. 021330.
To maintain the live BXSB.B6-Yaa+ colony (Stock No. 021330), BXSB/MpJ inbred females (Stock No. 000740) are bred with males hemizygous for the Y-linked C57BL/6-derived Yaa+ every generation.
