These Nlrp3 deficient mice may be useful for studying role of the inflammasome in immune response.

In this strain, a neo cassette replaces entire coding region of the NLR family, pyrin domain containing 3 (Nlrp3), abolishing expression. NLRP3 encodes the protein cryopyrin, which is a cytosolic nucleotide-binding domain and leucine-rich repeat containing (NLR) protein expressed in white blood cells and chondrocytes. Cryopyrin is involved in the formation of the inflammasome which regulates the immune system's response to injury, toxins, and infection by cleaving interleukin (IL)-1β. NLRP3 mutations are known to cause autoinflammatory diseases known as cryopyrin-associated periodic syndromes (CAPS) such as MWS, familial cold autoinflammatory syndrome (FCAS), and neonatal onset multisystem inflammatory disease (NOMID). Homozygous Nlrp3-deficient mice are viable and fertile. These mice exhibit a decrease in lactate dehydrogenase (LDH) in the bronchoalveolar lavage (BAL) fluids. Bone marrow derived macrophages from these Nlrp3-/- mice are not sensitive to the challenge by the bacteria peptidoglycan motif of muramyl dipeptide (MDP).
In an attempt to offer alleles on well-characterized or multiple genetic backgrounds, alleles are frequently moved to a genetic background different from that on which an allele was first characterized. We may modify the strain description if necessary as published results become available.
A targeting vector was designed to replace entire coding region of the NLR family, pyrin domain containing 3 (Nlrp3) gene with a neomycin resistance (neo). The construct was electroporated into 129S6-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the resulting chimeric males were bred to 129S6/SvEvTac females. These mice were bred to C57BL/6J mice for at least 10 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
