As a result of activation of the NOTCH signaling pathway, the transcriptional regulator, CBF1 (C promoter binding factor 1), binds with the cleaved intracellular domain of Notch(NICD).  The NICD-CBF1 complex then acts with co-activator mastermind-like (MAML) to promote transcription of various target genes.  These transgenic mice express a nuclear-localized fluorescent protein, the YFP variant Venus, under the control of multiple CBF1 (C promoter binding factor 1) binding sites and the basal simian virus 40 (SV40) promoter (also called CBFRE).  YFP Venus expression is consistent with canonical (CBF1-mediated) Notch activity, acting as a single cell resolution Notch signaling reporter.  In early postimplantation embryonic development transgene expression is detected in the epiblast of the peri-implantation embryos (approximately E5.0-5.25), in the anterior epiblast of E6.5 embryos, in the headfolds and in the blood islands (maturing erythrocytes surrounded by endothelial cells) of approximately E8.0 embryos.  At mid-gestation, transgene expression is detected mostly in the vasculature: dorsal aortic endothelial cells, umbilical vessels, pharyngeal arch arteries, for example.  At age E14.5, transgene expression is detected in the retinal pigmented epithelium, cuboidal epithelium of the anteriour lens, lung mesenchymal cells, developing heart valve cells, and other vascular cells.  Postnatally, reporter expression is detected predominantly in the vasculature, as well as the lung interstitial mesenchymal cells, kidney, adrenal gland and cardiac and skeletal muscle cells.  The transgene is expressed in the cerebellum, brain stem, telencephalon, mesencephalon, olfactory bulbs, and ganglia.  On this mixed STOCK background, homozygotes are viable and fertile.

 This CBF:H2B-Venus transgenic strain serves as a single cell resolution Notch signaling reporter and has applications in non-invasive and quantitative live imaging of canonical (CBF1-mediated) Notch signaling activity.
