EP1R TBC mice carry a targeted mutation of the Ptger1 gene with a loxP site flanked transcriptional blocking cassette preventing transcription of Ptger1. Cre recombinase mediated removal of the STOP cassette restores endogenous Ptger1 expression. This strain suitable for use in studies of inflammatory induced aversion, pain and malaise.

The Ptger1 gene encodes one of the four prostaglandin E2 receptors.
These mice possess a loxP site flanked transcriptional blocking cassette (TBC) inserted into the first intron of the targeted Ptger1 gene. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have the transcriptional blocking cassette deleted and restoration of Ptger1 expression in the cre-expressing tissues. Mice that are homozygous for this allele are viable and fertile.
When bred to a strain with Cre recombinase expression in the female germ line (see Stock No. 003651 for example), no transcript is detected in the renal collecting ducts by in situ hybridization in the resulting offspring. No
transcript is detected in the absence of Cre recombinase expression.
A targeting vector containing loxP site flanked was used to insert the floxed transcriptional blocking cassette into the first intron of Ptger1, 497 bp upstream of the start
codon. The construct was electroporated into 129S4/SvJae derived J1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. After testing for germline transmission, the resulting chimeric animals were bred to C57BL/6J mice.
Upon arrival, sperm was cryopreserved.
