These transgenic mice express full-length mouse Cd1d1 (CD1d1 antigen) cDNA under the regulatory control of the Lck (lymphocyte protein tyrosine kinase) promoter. The transgene is faithfully expressed in the T cell lineage (reported fold increase in thymus expression: CD4+(6x), CD8+ (4x), CD4+CD8+ (33x), CD4-CD8- (36x); in spleen CD4+ (4x), CD8+ (7.5x)).
Crosses with Cd1d1/Cd1d2 knockout mice to generate compound mutant lines were done to demonstrate that cortical thymocytes provide cell type-specific signals that lead to NKT cell differentiation.
A transgenic vector was designed to carry full-length mouse Cd1d1 cDNA inserted into a rabbit β-globin cassette under the regulatory control of the proximal Lck promoter. The transgenic vector was introduced to C57BL/6 embryos. Resultant mice were crossed with C57BL/6 background Cd1d1 knockout mice. The Cd1d1 knockout allele was later bred away leaving only the transgene. This strain was maintained on a C57BL/6 genetic background by the donating laboratory. This line corresponds to the Lck/low founder reported in Wei et al., J. EXP. MED. 2005, 202: 239-48.
