Description

Mapk1^flox (Erk2^fl) mice may be useful for conditional deletion of MAPK1 in tissues involved in development.

Detailed description

These Mapk1^flox/flox mutant mice possess loxP sites flanking exon 2 of the mitogen-activated protein kinase 1 (Mapk1) gene. MAPK1, also known as ERK2, is part of the Ras-Raf-ERK pathway, plays a role in both the initiation and regulation of meiosis, mitosis, and postmitotic functions in differentiating cells. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in cre-expressing tissues.

For example, when crossed to FVB-Tg(GFAP-cre)25Mes/J mice (Stock No. 004600) expressing Cre recombinase in the central nervous system, resulting mice behave abnormally and exhibit defect in neuron differentiation and cerebral cortex morphology.

Development

A targeting vector was designed to insert a loxP-flanked neomycin resistance (neo) cassette upstream of exon 2, and a single loxP site downstream of exon 2 of the mitogen-activated protein kinase 1 (Mapk1) gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a Cre expression plasmid to delete the neo cassette. Resulting ES cells contained multiple gene rearrangements; intact floxed-exon 2, intact floxed-neo cassette, or excision of both exon 2 and the neo cassette. Correctly targeted ES cells, containing only the floxed-exon 2, were injected into C57BL/6J blastocysts and resulting chimeric males were bred with 129 females. Resulting offspring were bred to establish a colony of Mapk1^flox/flox mice. These mice were bred to C57BL/6J mice for at least 11 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
