Detailed description

These floxed-SK3 mutant mice possess loxP sites flanking the translation initiation codon, coding sequences of exon 1, and a portion of intron1 of the potassium intermediate/small conductance calcium-activated channel, subfamily N, member 3 (Kcnn3) gene. SK3 is expressed in the soma and dendrites of dopaminergic neurons in the substantia nigra and influences their action potential frequency. Defects in dopamine (DA) releasing neurons have been sighted in pathologies such as schizophrenia and Parkinson's disease. SK3 is also expressed in smooth muscle of the bladder and uterus, and endothelia of the vasculature where the channels participate in blood pressure regulation. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 1 deleted in cre-expressing tissues.

Development

A targeting vector was designed to insert a loxP site upstream of the translation initiation codon in exon 1 of the potassium intermediate/small conductance calcium-activated channel, subfamily N, member 3 (Kcnn3) gene. A frt-flanked neomycin resistance (neo) cassette and a second loxP site were inserted downstream of exon 1. The construct was electroporated into 129S6/SvEvTac-derived Duffy embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric males were bred to C57BL/6J females. Offspring, heterozygous for this SK3 allele, were bred with Flp transgenic mice to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. These mice were bred to C57BL/6J mice for at least 15 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
