These Ago1fl/fl mice possess loxP sites flanking a portion of the coding region of the eukaryotic translation initiation factor 2C, 1 ( Eif2c1) gene and have applications in studies related to post-transcriptional gene regulation and the microRNA (miRNA) pathway.

The Argonaute 1 protein, encoded by Eif2c1 (eukaryotic translation initiation factor 2C, 1) is essention to RNA-mediated gene silencing: it binds to both miRNAs and siRNAs, inhibiting mRNA translation. These mice possess loxP sites flanking exons 13 through 16 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these knockout mice are bred to mice that express Cre recombinase, resulting offspring will have exons 13 through 16 deleted in the cre-expressing tissues.
Dr. Alexander Tarakhovsky (while at The Rockefeller University) generated a targeting vector containing FRT site flanked NEO cassette and 2 loxP sites that was used to flank exons 13-16 with loxP sites. This construct was electroporated into 129P2/OlaHsd derived E14.1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to FLP-expressing mice on the C57BL/6 genetic background (FLPeR) to remove the FRT site flanked NEO cassette. Mice that retained loxP site flanked exons 13-16 were then bred to other transgenic mice on the CD1 genetic background. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to remove the transgenes and establish the colony.
