Molecular Note | A construct was inserted into the locus that contained the following elements (5' to 3'): the tetracycline transactivator (tTA), two poly-adenylation sequences, the tetracycline operator with minimal CMV promoter, a floxed neo cassette with 4 poly-adenylation sequences, the mouse transcript, and a poly-adenylation sequence. The locus promoter drives expression of tTA while the tet operator drives expression of the transcript only after cre mediated removal of the floxed neo-STOP cassette that can be repressed by doxycycline treatment. |
