Description

In this strain a CDX2-EGFP fusion protein is expressed under the control of the endogenous Cdx2 promoter. Green Fluorescent Protein fluorescence mimics endogenous Cdx2 expression and is detected in both live and fixed embryos. This mutant mouse strain may be useful lineage tracing and live imaging of pre-implantation embryos.

Detailed description

Cdx2, caudal type homeobox 2, a transcription factor essential in mice for trophoblastic and placental development, is also a marker for intestinal epithelial development and colon cancer. These mice carry a targeted mutation in which EGFP, Enhanced Green Fluorescent Protein, has been inserted in-frame in exon 3. A CDX2-EGFP fusion protein is expressed under the control of the endogenous Cdx2 promoter. Mice that are homozygous for the targeted mutation are viable and fertile. Expression of the fusion protein is detected in pre-implantation stages, the primitive streak of 7.0-7.5 d. p. c. embryos, and the epithelial layer of intestinal villi of adult mutants. EGFP expression mimics endogenous Cdx2 expression, and fluorescence is detected in live and fixed tissues.

Development

The RP23-7616 mouse bacterial artificial chromosome (BAC) containing the Cdx2 gene was modified by inserting sequence encoding EGFP into exon 3 of the Cdx2 gene. DNA from the modified BAC was used in the targeting construct that was electroporated into C57BL/6 X 129Sv F1 hybrid derived ESV 6.5 F1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into CD1 blastocysts. The resulting chimeric animals were crossed to CD1 and 129SvEv mice. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
