Description

These transgenic mice express an inducible version of cre recombinase mice under the direction of a Sox9 promoter. They are suitable for performing cre-recombination in pancreatic ductal cells and their progenitors.

Detailed description

Mice hemizygous for the BAC Sox9-creER^T2 transgene are viable and fertile. Cre-ER^T2 expression is directed by the murine SRY-box containing gene 9 (Sox9) promoter/enhancer regions in early pancreatic progenitor and ductal cells. Restricted to the cytoplasm, Cre-ER^T2 can only gain access to the nuclear compartment after exposure to tamoxifen. When these mice are bred with mice containing loxP-flanked sequence, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the floxed sequences in the cre-expressing cells of the offspring.

For example, when bred to B6.129X1-Gt(ROSA)26Sor^tm1(EYFP)Cos/J mice (Stock No. 006148), tamoxifen induction in the offspring results in fluorescent labeling of pancreatic progenitor cells such as endocrine, acinar, and duct cells, until birth, with weak fluorescence in non-β endocrine cells for 3 weeks after birth. Fluorescence is also seen in 70% of pancreatic ductal cells in adults.

Development

Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23-229L12) containing the entire mouse Sox9 (SRY-box containing gene 9) gene. This BAC was modified to contain a KOZAK-CreER^T2(a Cre recombinase fused to a human estrogen receptor ligand binding domain)-polyadenylation sequence in place of the Sox9 start codon. This BAC also contained a β-galactosidase (LacZ) gene that the donating investigator reported to be inactive. The modified BAC was microinjected into fertilized (BALB/c x C57BL/6)F2 oocytes and the resulting offspring were bred to CD1 outbred mice. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
