These mice carry a conditional floxed allele of the Actl6a gene that affects DNA conformation and influences the transcriptional activation or repression of genes.

A loxP-flanked neomycin resistance cassette with a thymidine kinase (TK) promoter was inserted between both exon 5 and 6, and exons 3 and 4 using 129S6/SvEvTac-derived TC1 embryonic stem (ES) cells. The neomycin cassette in between exons 5 and 6 was excised through crosses with a Cre strain, leaving a single loxP site in intron 5 and a loxP-flanked neomycin cassette in intron 3. This strain was backcrossed to C57BL/6 for at least 5 generations by the donating lab.
