RAPGEF3 (or EPAC1) is a rap guanine nucleotide exchange factor that functions as an intracellular receptor for cyclic AMP. Epac1 mRNA exhibits widespread expression. These mutant mice possess loxP sites flanking exons 3 to 6 of the (Rapgef3) gene. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express Cre recombinase, the resulting offspring will have exons 3-6 deleted in the cre-expressing tissues. When a global Rapgef3 knockout is combined with Rapgef4Gt(OST450264) (Epac2 KO) (Stock No. 018390), double homozygous knockout mice exhibit impaired long term potentiation (LTP), deficits in spatial learning, memory deficits, and abnormal social interactions.
A targeting vector was designed to insert a loxP site upstream of exon 3 followed by a frt-flanked neomycin resistance (neo) cassette, and a second loxP site downstream of exon 6. The construct was electroporated into 129S2/SvPas-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to C57BL/6 mice. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
