These Agtfl mutant mice possess loxP sites flanking exon 2 of the angiotensinogen (Agt) gene. AGT is a circulating α-2-globulin produced mainly by the liver. It is the only known precursor to the peptide angiotensin II (ANG II), which is a hormone linked to hypertension. Mice that are homozygous for this allele are viable, fertile, normal in size and do not display any gross physical or behavioral abnormalities. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues.
For example when bred to B6.Cg-Tg(Fabp4-cre)1Rev/J mice (Stock No. 005069), AGT depletion in adipocytes results in decreased systolic blood pressure and reduced circulating AGT concentrations.
A targeting vector was designed to insert a loxP site upstream of exon 2 and a floxed and frt-flanked neomycin resistance (neo) cassette downstream of exon 2 of the angiotensinogen (Agt) gene. The construct was electroporated into C57BL/6NTac-derived iTL C57/Bl6 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric mice were bred together. The resulting offspring were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J transgenic mice (Stock No. 003800) to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. These Agtfl mice were bred to C57BL/6 mice for at least 3 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
|Allele Name||targeted mutation 1.1, inGenious Targeting Laboratory|
|Allele Type||Targeted (Conditional ready (e.g. floxed), No functional change)|
|Gene Symbol and Name||Agt, angiotensinogen (serpin peptidase inhibitor, clade A, member 8)|
|Strain of Origin||C57BL/6NTac|
|Molecular Note||A loxP site was inserted upstream of exon 2 and an FRT and loxP flanked neo cassette were inserted downstream of exon 2 via homologous recombination. Flp mediated recombination deleted the neo cassette leaving exon 2 floxed.|
When maintaining a live colony, homozygous mice may be bred together.
