These Ror1f/f conditional mutant mice may have applications in studies related to Wnt5a-mediated signaling during embryonic development.

These Ror1f/f mutant mice possess loxP sites flanking exons 3-4 of the receptor tyrosine kinase-like orphan receptor 1 (Ror1) gene. Mice that are homozygous for this allele are viable and fertile. ROR1 is a receptor for Wnt5a-mediated signaling. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 3-4 deleted in the cre-expressing tissues.
When bred to Tg(EIIa-cre)C5379Lmgd transgenic mice, ROR1 null mice are morphologically and phenotypically normal, although some exencephaly has been seen. When Ror2 gene expression is also abolished (see Stock No. 018354) these double KO mice exhibit decreased branching of the sympathetic neuron axons, facial malformations, and truncation of the limbs and posterior region. Double KO mice die by E14.5.
A targeting vector was designed to insert a loxP site upstream of exon 3 followed by a frt-flanked neomycin resistance (neo) cassette, and a second loxP site downstream of exon 4 of the receptor tyrosine kinase-like orphan receptor 1 (Ror1) gene. The construct was electroporated into 129S4/SvJae-derived J1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric males were bred to C57BL/6 females. Offspring were bred with B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ transgenic mice (Stock No. 009086) to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. These mice were maintained on a mixed B6x129 background. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
